Putnam Valley, NY. (Apr. 23, 2014) People who have had a stroke, often suffer motor deficits with little potential to restore neurological function. However, a study conducted in Taiwan, that will be published in a future issue of Cell Transplantation, but is currently freely available on-line as an unedited early e-pub at: http://www.ingentaconnect.com/content/cog/ct/pre-prints/content-ct1168Chen, has found that when one group of stroke victims had their own peripheral blood stem cells (PBSCs) injected directly into the brain and a similar group did not, those who received the PBSCs experienced some "improvement in stroke scales and functional outcome." Those in the PBSC-injected group also received injections of the growth factor granulocyte-colony stimulating factor (G-CSF), known to be potentially neuroprotective.

"In this phase 2 study, we provide the first evidence that intracerebral injection of autologous (self-donated) PBSCs can improve motor function in those who have suffered a stroke and have motor deficits as a result," said study corresponding Dr. Woei-Cheng Shyu of the Center for Neuropsychiatry, Graduate Institute of Immunology and Translational Medicine Research Center, China Medical University in Taiwan. "Our study demonstrated that this therapeutic strategy was feasible and safe in stroke patients who suffered a prior stroke, but within five years from the onset of symptoms."

According to the authors, there has been little advance made in restoring neurological function following ischemic stroke. However, since neuronal death is the primary mechanism that limits functional recovery, stem cell therapy is emerging as a potentially effective regenerative approach. Once more PBSCs are being increasingly used as a self-donated source for cell therapies for regenerating skeletal muscle, heart and neurons. The PBSCs may need to be "amplified" with G-CSF, speculated the researchers.

All of the patients in the trial had suffered a stroke in the past, as long as five years prior to this study. At the end of a 12 month follow-up, the group of 15 patients with neurological deficits who received injections of PBSCs experienced neurological and functional improvement based on a number of clinical outcomes measures. The control group of 15 patients with neurological deficits that did not receive the PBSC injections did not experience the same beneficial outcomes.

The researchers reported that nine of the 15 patients undergoing PBSC transplantation experienced "positive motor evoked potentials" (MEPs) after transcranial magnetic stimulation, but why MEPs appeared in some of the transplanted group, but not all, was unclear.

"Despite this success, it should be noted that this was a preliminary study and, due to the small number of patients, are tentative," concluded the researchers. "In the future we plan to conduct a multi-center, large-scale, double blind, placebo-controlled randomized studies to better evaluate the effect of PBSC implantation in patients suffering from the effects of past stroke."

"This phase II study offers pilot clinical evidence supporting the use of autologous stem cell-based treatment for stroke" said Dr. Cesar V. Borlongan, Prof. of Neurosurgery and Director of the Center of Excellence for Aging & Brain Repair at the University of South Florida. "Clarification of the impact of G-CSF on the cells and whether other factors are necessary to maximize the benefit of cell transplantation, as well as further studies with a larger number of patients are necessary to determine equivocal safety and efficacy of this treatment".

Orbsen Therapeutics, a spin-out from NUI Galway's Regenerative Medicine Institute (REMEDI), is to partner with the University of Birmingham in a E6 million EU FP7 funded MERLIN project to fight liver disease.

The EU FP7-funded project known by the acronym "MERLIN" (MEsynchymal stem cells to Reduce Liver INflammation) is led by Professor Phil Newsome, Clinical Director of the Birmingham University Stem Cell Centre. MERLIN will advance Orbsen's proprietary cell therapy to a Phase 2a clinical trial in patients with inflammatory liver disease. This MERLIN project will evaluate the Orbsen cell therapy in 4 different research laboratories across Europe and the project will culminate in a Phase 2a clinical trial of the therapy in the crippling inflammatory liver disease, Primary Sclerosing Cholangitis.

This is Orbsen's fourth success in attracting FP7 funding (the EU's Seventh Framework Programme for Research), making them one of Ireland's most successful private companies in this funding programme and now connects Orbsen to 23 global collaborators. Other successful cell therapy projects for Orbsen include PURSTEM (completed), REDDSTAR (ongoing) and DeCIDE (ongoing).

Orbsen Therapeutics Ltd. is a privately-held company founded in 2006 as a spin-out from Ireland's Regenerative Medicine Institute (REMEDI) in NUI Galway. As part of the PurStem EU FP7 program, Orbsen developed proprietary technologies that enable the prospective purification of highly defined and therapeutic (stromal) cells from several human tissues, including bone marrow, adipose tissue and umbilical cord.

Orbsen's CEO Brian Molloy said, "Orbsen has secured substantial amounts of research funding in the last 18 months which will further validate our product and bring us through to a "first in man" clinical trial in 2015/16. Our model has always focused on putting the 'science first' and we have successfully used that approach to develop a technology that could potentially position us and indeed Ireland at the leading edge of European Cell Therapy development."

Mr Molloy continued, "As a spin-out from the NUI Galway based REMEDI Institute we have focused the majority of our collaborations with an Irish research team. Our success in the MERLIN project now demonstrates that we are capable of playing a key role in collaborations led by researchers across Europe."

The total research budget for the MERLIN project is close to E6 Million of which E1 Million will go directly to Orbsen Therapeutics over the 4-year period of the project.

Two Central Pennsylvania dogs are receiving a regenerative therapy for arthritis thats unprecedented for this area and less expensive than standard surgery. Stem Cell therapy is a way to repair damaged tissue and treat injury. When dealing with dogs, veterinarians say its the future of treatments and its becoming less costly.

Gunny is a 7-year-old German Shepard. He underwent the revolutionary stem cell therapy at the Palmyra Animal Clinic. Vets say the stem cell therapy is a way to combat Gunnys arthritis in his hips. Doctors collected fatty tissue from his shoulder, processed the stem cells in the lab and injected the cells back into his hips. This happens all in one day for around $1500. Prior to this, surgery could cost around $3,000.

Dr. Calvin Clements of the Palmyra Animal Clinic says, Injected in a damaged joint or ligament, these cells will take on that characteristic and differentiate into the cartilage or tissue were dealing with and help to regenerate it.

Dr. Clements says results are noticeable in about a month. On average, animals improve 85%.

Gloria Arroyos stem cell therapy doc blamed for cancer patient’s death

A doctor offering stem cell therapy may face charges for the death of a cancer patient who allegedly underwent treatment similar to that administered to former president and incumbent Pampanga Rep. Gloria Macapagal-Arroyo.

This was after it was found out that she is not a licensed doctor in the Philippines.

A report on GMA News TV's "News To Go" on Wednesday said a complaint has been filed against Dr. Antonia Carandang-Park at the National Bureau of Investigation (NBI) by Bernard Tan, who claims that his daughter, Kate, died after going through the said alternative treatment.Cancer patient Kate Tan received stem cell therapy from Dr. Antonia Carandang-Park, Gloria Arroyo's doctor.

Park owns the Tagaytay-based Green & Young Health and Wellness Center where Arroyoburdened by persistent trouble with her cervical spinesought treatment in 2012.

In an interview with GMA News, Tan said his daughter, who had Hodgkin's lymphoma (a type of cancer of the blood), was given "the same treatment that [Park] did with Gloria," which included "juicing diet, vegetable diet... acupuncture coffee enema, at 'yun na nga, stem cell."

Stem cell therapy introduces new adult stem cells into damaged tissue in order to treat disease or injury.

"Ang sabi niya, 'Give me three months, magaling na 'yan,'" Tan told GMA News. He added that his family was easily convinced to take their daughter to Park's wellness center because "Presidente na ng Pilipinas ang pumunta doon."

"Siguro naman na-scrutinize na nila 'yan, na-background check na nila 'yan," he said. "Kumbaga, 'yung credibility no'n, nag-build up na."

Kate was fed nothing but bananas and vegetable juices for three months, and had eight rounds of "embryonic" stem cell treatment, he said.

However, the 23-year-old lost even more weight, prompting the family to seek the assistance of a different doctor. Kate had eight rounds of 'embryonic' stem cell treatment, her father Bernard Tan said.Seven months later, in July 2013, Kate passed away.

Scientists at The University of Nottingham have developed a new substance which could simplify the manufacture of cell therapy in the pioneering world of regenerative medicine.

Cell therapy is an exciting and rapidly developing area of medicine in which stem cells have the potential to repair human tissue and maintain organ function in chronic disease and age-related illnesses. But a major problem with translating current successful research into actual products and treatments is how to mass-produce such a complex living material.

There are two distinct phases in the production of stem cell products; proliferation (making enough cells to form large tissue) and differentiation (turning the basic stem cells into functional cells). The material environment required for these two phases are different and up to now a single substance that does both jobs has not been available.

Now a multi-disciplinary team of researchers at Nottingham has created a new stem cell micro-environment which they have found has allowed both the self-renewal of cells and then their evolution into cardiomyocyte (heart) cells. The material is a hydrogel containing two polymers an alginate-rich environment which allows proliferation of cells with a simple chemical switch to render the environment collagen-rich when the cell population is large enough. This change triggers the next stage of cell growth when cells develop a specific purpose.

Major priority

Professor of Advanced Drug Delivery and Tissue Engineering, Kevin Shakesheff, said:

Our new combination of hydrogels is a first. It allows dense tissue structures to be produced from human pluripotent stem cells (HPSC) in a single step process never achieved before. The discovery has important implications for the future of manufacturing in regenerative medicine. This field of healthcare is a major priority for the UK and we are seeing increasing investment in future manufacturing processes to ensure we are ready to deliver real treatments to patients when HPSC products and treatments go to trial and become standard.

The research, Combined hydrogels that switch human pluripotent stem cells from self-renewal to differentiation, is published in the Proceedings of the National Academy of Sciences (PNAS).

Arroyo’s alternative medicine doctor unlicensed

Dr Antonia Park, according to the Professional Regulatory Commission, is not authorized to practice medicine in the Philippines. She faces charges of estafa and reckless imprudence resulting in homicide.

MANILA, Philippines The alternative medicine doctor who took in former president Gloria Macapagal-Arroyo in 2012 for possible stem cell therapy is not licensed to practice in the Philippines.

On Tuesday, April 1, Antonia Park of the Green & Young Health & Wellness Center admitted to Rappler in a phone interview she is not a registered physician in the Philippines because "I'm not from here." She is instead a registered medical practitioner in London and Korea.

"Thats why its a wellness center. If dito ako [registered], maglalagay na lang ako ng medical center," she said, referring to her center located in Tagaytay City. (That's why it's a wellness center. If I am registered here, I might as well put up a medical center.)

A document from the Professional Regulatory Commission (PRC) dated Aug 12, 2013 showed Park is not in the database of physicians which contains the names of those duly authorized to practice medicine in the Philippines.

Park and some of her clinic staff are facing charges of estafa and reckless imprudence resulting in homicide. The charges were filed last year by businessman Bernard Tan with the National Bureau of Investigation (NBI) after his 23-year-old daughter Kate died.

Kate, an Ateneo student who graduated with honors in March 2013, died 4 months later on July 4, due to a tumor that blocked the entry of blood to her heart, secondary to Hodgkin's Lymphoma. The disease is an uncommon but curable cancer of the lymphatic system.
